Does SYBR Safe require UV light?
We recommend that SYBR Safe DNA Gel Stain be protected from light during storage and gel staining. However, it is sufficiently stable to withstand UV illumination for >30 minutes; realistically, hours of constant UV or bright room light exposure are required to cause any significant loss of signal.
How does SYBR Green fluorescence?
Posted June 22, 2020. SYBR Green is one of the most commonly used fluorescent dyes in qPCR. It binds to double-stranded DNA molecules by intercalating between the DNA bases. Once intercalated to DNA, SYBR Green becomes less mobile, causing its energy to be released as fluorescence.
How does SYBR Green work in gel electrophoresis?
It binds specifically to double-stranded DNA. Using this dye, double-stranded DNA molecules can be exclusively quantified in the presence of single-stranded DNA molecules during denaturation experiments. SYBR Green I shows maximum fluorescence at excitation and emission wavelengths of 497 and 520 nm, respectively.
Is SYBR Green sensitive to light?
SYBR Green I dye (present in the master mix) is light-sensitive; solutions containing the master mix should be protected from light whenever possible. Note Total reaction volumes of 50 μl may also be used.
