What does a Ponceau stain tell you?
Ponceau S stain is useful to locate and identify the proteins transferred electrophoretically onto nitrocellulose/nylon membranes before the antibody-mediated detection as the dye does not interfere with antibody detection in Western blots.
How do you remove Ponceau stains?
Completely submerge with Ponceau stain and put on rocker ▪ Nitrocellulose membranes saturate in ~5 min. PVDF membranes saturate in ~15 minutes. 3. Remove Ponceau from non-protein parts of the membrane via gentle rinsing with distilled H2O.
Can you Ponceau stain after stripping?

How do you use Ponceau?
Transfer membrane to 5ml Ponceau S stain solution on an orbital shaker for 5 minutes at RT. Rinse membrane with DI water to achieve desired staining, approximately 2-3 washes of 5 minutes each. For protein destaining, wash the membrane with 0.1N NaOH solution for 5 minutes and repeat once.
